The Church of the Holy Trinity is one of the oldest churches in Odessa. In 1795, Metropolitan Gabriel (Banulesko-Bodoni) of Ekaterinoslav and Tauris laid the first four churches, one of them «in the name of Holy Trinity for the dissenters of the Greek nation on the Greek forshtat».
Initially it was a wooden church, and on June 29, 1804 the solemn laying of the stone church took place. The temple was built according to the project of architect F. Frapoli and in the spring of 1808 was solemnly consecrated.
The first rector of the Holy Trinity (Greek) temple was Archpriest John Rodesu. In 1838 by his care the church was expanded by two limits, which acquired the final cross-shaped form in the Byzantine style with one dome and a bell tower with a spire. The right side of the church on March 31, 1840 was consecrated in memory of the Annunciation of the Blessed Virgin Mary.

The side limits have undergone changes in our time: the southern limit is named in honor of the Protection of the Most Holy Mother of God, and the northern limit is named in honor of the icon of the Mother of God, called «Joy Unexpected», and St. Spyridon of Tremifunda, the Wonderworker.
In the early XIX century, the Greek Church of the Holy Trinity was the spiritual center of the Greeks — patriots. The Greeks of Odessa fervently supported the struggle for independence of the people of Greece. The rebellion that began in 1821, led to the fact that the Turkish authorities executed the Ecumenical Patriarch of Constantinople Gregory V, fearing his influence on the rebellious Greeks. Thrown into the sea, the body of the patriarch was discovered by Russian sailors and brought to Odessa. The remains of the martyred patriarch were buried in the Holy Trinity (Greek) Church.

In addition, in and near the temple were buried Greek archpriests who died at different times. In May 1907 at the eastern wall of the temple in a special annex was buried philanthropist and public figure, one of the most prominent members of the Greek community, honorary citizen of Odessa Grigory Grigorievich Marazli.
In 1900-1908 the temple was reconstructed under the direction of architect A. Todorov.
During the Bolshevik persecution of the Church, the temple was closed, its premises were turned into a gymnasium, numerous burials were looted.
After the Great Patriotic War, a new stage in the history of the Holy Trinity (Greek) Church begins. The church was decapitated (without the dome and bell tower), there was a lot to do to restore it.

In 1954, a sub-church of the Patriarch of Alexandria in the Russian Orthodox Church was established at Holy Trinity (Greek) Church. According to the approved regulations, the services were conducted in Greek and Church Slavic languages, and all exarchs, at the same time, were citizens of Greece.
The Alexandrian Orthodox Church sub-church existed until April 1, 1999, after which it was transferred from Odessa to Moscow, and the Holy Trinity (Greek) Church was returned to the status of a city church of the Odessa diocese of the Ukrainian Orthodox Church.
In the mid-90s of the XX century over the temple shone a new dome and a slender bell tower, restoration work continues.
In the temple there is a venerable image of the Mother of God «Joy in Need», before which according to the established tradition every Friday evening the akathist to the Most Holy Mother of God is performed. In the temple there are Sunday schools for adults and children. There is a parish library.
